RESEARCH EXPERIENCE:
ACADEMIC PROJECT :
As a part of my Academic curriculum in M.sc I conducted a research work entitled "Isolation, characterization and Identification of Leuconostoc mesenteroides From foods, water and woman".The core objective of the dissertation is to find the solution for those lacunae encountered in the research work on Leuconostoc mesenteroides. The present work aimed at Isolation, characterization, Identification, to determine the mode of transfer , the sources of inoculum and to determine transfer of personnal inoculation involved in idli dough prepration. To clear and verify the substantial lacuna on motility aspect.
In FOOD MICROBIOLOGY Leuconostoc mesenteroides function in idli fermentation and sauerkraut production. The degree of importance in INDUSTRIAL MICROBIOLOGY causes various disorders that reduces qualitative and quantitative product of sugar in sugar refineries, that leads to 1% LOSS OF SUGAR in overall Industrial sugar production, by conversion of sugar into DEXTRAN by dextran sucrase enzyme produced by Leuconostoc mesenteroides, Reference "Cauvery sugars and chemicals limited Tamilnadu". In MEDICAL MICROBIOLOGY it can be isolated from human infections cases such as meningities, endocardites , septicemia and in immunosuppresed patients.
Samples such as Ricegram dough, soaked rice water, sugarcane juice, sauerkraut juice, skins scrapings from personnal involve in idli dough prepration and cultivated in BASAL MEDIUM Nutrient Agar, from which Leuconostoc sp selected in SELECTIVE MEDIUM Vancomycin Agar and was confirmed by ENRICHED MEDIUM AND DIFFERENTIAL MEDIUM Sheep Blood Agar ( No Hemolysis) and Chocolate Agar ( Non Fastidious) and Vancomycin [30 mic g] Disk Diffusion tested in Muller Hinton Agar to confirm Vancomycin resistance. To identify species it was inoculated in to Sucrose Agar to detremine Dextran production characterized by slimmy gummy appearances of colonies produced by Leuconostoc mesenteroides. Isolates from idli dough, sugararcane, soaked rice and personnal skin scrapping[ hand] were found to be Dextran producing Leuconostoc mesenteroides. Isolate from idli dough were subjected to Biochemical test and Physiological test. The strain was standardized.
AIM AND SCOPE
An approach was made to determine how the mode of transfer of Leuconostoc mesenteroides by water, grams, cabbage leaf, and food handlers were found to be sources of inoculum. Another apporoch was made to clear the lacuna regarding motility exibited by Leuconostoc mesenteroides it was found to be motile vide Hanging drop technique, diffusion in SSNA and ILMA. BERGES MANUL declares that all the species of Leuconostoc were Non motile but TOPPLEY WILLSON says that some of the Leuconostoc species can exibit motility. Which was confirmed in this dissertation that Leuconostoc mesenteroides were motile. This dissertataion contribute newer information regarding the presence of Leuconostoc mesenteroides in Traditional south Indian food ‘soaked rice’as fermentor. The dissertation supports the supply of Inoculum for idli dough fermentation in cold countries, hill’s region and even in winter season .To maintain the inoculam a maintenance media was recommended . To over come the industrial loss of sugar caused by Leuconostoc mesenteroides biocide also recommended in this dissertation. Scanning Electron Microscopic Structure studied to determine the ultramicroscopic details of Leuconostoc mesenteroides that reports for motility result awaited. On futurological aspect the study stronglly recommend to look for SCP PRODUCTION in Leuconostoc mesenteroides and molecular biology study.
ACADEMIC PROGRAMME
Presented paper in " XXIV NATIONAL CONGRESS OF INDIAN ASSOCIATION OF MEDICAL MICROBIOLOGIST" from 17th Nov to 19th Nov 2K held at Belgaum JN Medical college Karnataka, India. AB.NO.BO68. "A STUDY ON THE IN VITRO ANTIBACTERIAL PROPERTIES OF OCIMUM BASILICUM LINN VAR. sTHYRSIFLORUM LEAF EXTACT (AQUEOUS) FOR GASTROENTERITIC AND PYOGENIC BACTERIA."
ABSTRACT: Ocimum Basilicum linn var. Thyrsiflorum (White Basil), is a common sweet basil, claimed to cure pimples and other skin disorders, diarrhoea & dysentery. Review of literatute reveals inconsistency in the results obtained on the antibacterial properties of the volatile oil from the leaves of white basil. As siddha practitioners use aqueous extract , seitz filterate of the fresh leaf extract was used to know its antibacterial effect on certain gastro-enteritic bacteria (Salmonella typhi, Salmonella paratyphi, Escherichia coli, Shigella dysenteriae, Vibrio cholerae, Vibrio parahaemolyticus) and pyogenic bacteria (Staphylococcus aureus, Streptococcus pyogenes and Serratia marcescens). The seitz filtered extract of white basil was mixed into the medium at three concentrations (1.71 mg/ml, 3.2 mg/ml & 6.0 mg/ml). The growth of Vibrio cholerae and Vibio parahaemolyticus were markedly inhibited. Salmonella typhi and Salamonella parathyphi were slightly inhibited. However the pyogenic bacteria were not inhibited. Ocimum basilicumLinn var thyrsiflorum leaf extract seems to exhibit appreciable inhibitory effect for Vibrio spp. And lack of inhibitory effect for pyogenic bacteria
Presented paper in " XXI ANNUAL CONFERENCE INDIAN ASSOCIATION OF BIO-MEDICAL SCIENTIST " Held at Raja Muthiah Medical College, Annamali University, India. AB.NO.78. PAPER ENTITLED "A STUDY ON THE IN VITRO ANTIFUNGAL PROPERTIES OF AQUEOUS EXTRACTS FROM FLOWER BUDS, SEEDS AND BARK FROM CASSIA AURICULATE LINN. FOR OPPORTUNISTIC AND DERMATOPHYTIC FUNGI".
ABSTRACT: Leafs, Flower, Buds, Roots And Pods OF tanners cassia (Cassia auriculate) has been used in the preparation of sidha medicine, (Avaram Choornam). The present work was aimed to evaluate the in vitro antifungal preparation of flower buds, bark and seeds of the plant for three Dermatophytes. Trichophyton mentagrophytes, gypseum and Epidermataphyton floccossum and give opportunistic fungi (Candida) albicans, Aspergillus flavus, pencillium,Rhizopus sp and mucor species. Sterile & aequeous extracts of the plant parts were prepared and each extract was mixed into S.D.A medium to obtain extract medium in three different concentrations 10%, 20%, 30%, With control. The growth of all three dermatophytes were completely inhibited by the extract even at 10% concentration. The results of the inhibitory effect of three extract on opportunistic fungi varied. The flower bud extract seemed to posses the highest inhibitory on the dermataophytic & opportunistic fungi.
